Scientific Overview

The hydrophobic internal cavity of gamma cyclodextrin enables it to form host-guest inclusion complexes with a wide range of lipophilic drug molecules, improving their aqueous solubility, chemical stability, and dissolution rate. Its larger cavity size compared to alpha- and beta-cyclodextrin makes it particularly suited to encapsulating bulkier drug molecules, and it is notably the parent structure used to manufacture sugammadex.

Gamma cyclodextrin is a cyclic oligosaccharide composed of eight glucose units linked by a-1,4-glycosidic bonds, forming a truncated cone-shaped molecule with a hydrophilic exterior and hydrophobic cavity. It is widely used in pharmaceutical formulation to solubilize, stabilize, and enhance the bioavailability of poorly water-soluble drug molecules.

Peer Queries & FAQs
Why is gamma cyclodextrin used over alpha or beta cyclodextrin?
Its larger 8-glucose-unit cavity accommodates bulkier guest molecules and offers higher water solubility with a favorable renal safety profile, making it well-suited for parenteral use.
What drug products use gamma cyclodextrin?
It forms the core structure of sugammadex (neuromuscular block reversal agent) and is used as a solubilizing excipient in various IV formulations.
Is gamma cyclodextrin safe for injectable use?
Yes, it is listed in the FDA Inactive Ingredient Database for IV injection formulations and is generally recognized as safe (GRAS).
What is the recommended storage condition?
Store in a tightly sealed container at room temperature, protected from moisture, as cyclodextrins are hygroscopic.
